摘要： Finger millet (Eleusine coracana L. Gaertn.) is a nutritious and climate-resilient crop with a C-4 type carbon fixation pathway. The present study was aimed to assess the drought tolerance capacities of four finger millet genotypes based on their physiological and biochemical characteristics at three different phenological stages. Finger millet genotypes RAU 8, GPU 67, GPU 28 and MS 9272 were subjected to two water regimes, regular irrigation (control) and suspended irrigation (drought stress). During water regimes, morpho-physiological [biomass accumulation, leaf relative water content, and photosynthetic pigments] and biochemical changes [proline content, water soluble carbohydrates, antioxidant enzymes, and malondialdehyde content] were studied during seedling (18th day), vegetative (49th day) and early flowering stages (73rd day). The maintenance of growth especially root growth, biomass accumulation, the differential response in the concentration and changes of pigments, accumulation of proline, water-soluble carbohydrates and increased levels of antioxidant enzymes under drought stress play a major role in differential tolerance in finger millet genotypes that is conferred by the biplot analysis. The genotype RAU 8 is the most drought-tolerant genotype at all the three different phenological stages. Whereas the genotype GPU 67 was identified as sensitive at the seedling stage and its tolerance level was improved at vegetative and early flowering stages. The genotypes GPU 28 and MS 9272 were considered as drought sensitive at all three different phenological stages. Our results provide inputs to the breeders to select genotypes as parents and to design effective strategies in crop improvement programs.

摘要： This experiment was undertaken to evaluate the effects of different levels of soluble and insoluble fibers on growth performance and blood indices in ostrich at the age of 1 to 6 months. Thirty 30-day-old ostriches (males and females) were randomly assigned to five treatments with six replicates. There were five dietary treatments consisted of (1) basal diet;(2 and 3) diets containing 2 and 4% more soluble fibers (SF) than basal diet, and (4 and 5) diets containing 2 and 4% more insoluble fibers (ISF) than the basal diet. The dietary treatments had no significant influence on growth performance of ostriches. By increasing the levels of SF and ISF, the concentration of blood glucose (P < 0.0001) and high-density lipoprotein (HDL,P = 0.0046) increased, while the plasma concentration of triglyceride (P < 0.0001) and very-low-density lipoprotein (VLDL,P = 0.0006) decreased. The plasma enzyme activity of alanine aminotransferase (ALT) decreased in ostriches fed diet containing 4% more SF as compared with control (P < 0.0006). Body weight gain (BWG) at the starter phase (3-4 months) was higher than the pre-starter (1-2 months) and grower (5-6 months) phases (P < 0.0001). The highest concentration of blood glucose was observed at 2 months of age while the lowest magnitude was found at 4 months of age (P < 0.0001). The plasma cholesterol concentration increased at 4 months and decreased at 6 months as compared with 2 months (P < 0.0001). The concentration of HDL and LDL at 4 months of age was higher than two other ages (P < 0.0001). The blood concentration of triglyceride (P < 0.0001), VLDL (P < 0.0001), and ALT (P = 0.0005) decreased as ostriches grew up. It could be concluded that increasing the levels of soluble and insoluble fibers in the diet up to 4% more than previous recommendation may not have negative effects on growth performances and blood indices in ostrich.

摘要： Background Ursodeoxycholic acid (UDCA) has been widely recommended as the first-line drug for primary biliary cholangitis (PBC) in the current guidelines. However, its therapeutic effects are poor in nearly one-third of patients. The early identification and intervention of these patients is crucial for delaying disease progression. Therefore, we explored risk factors for inadequate biochemical response and constructed a nomogram to predict the potential risk. Methods We enrolled 356 patients and randomly divided them into training (70%) and validation groups (30%). We defined inadequate biochemical response as the study endpoint. Logistic analysis was used to identify the independent predictors of poor biochemical response. Based on these factors, a predictive nomogram was finally constructed. Then, discrimination and calibration were evaluated by internal validation. Additionally, the association between the model predictions and prognosis was further analyzed. Results Female sex, and albumin and bilirubin concentrations were identified as risk factors, and a nomogram was built based on these factors. The areas under the ROC curves of the training and validation groups were 0.809 and 0.791, respectively. Moreover, calibration curves showed that predictions of the nomogram had good concordance with the actual outcomes. The correlation analysis demonstrated that PBC patients with a high probability of a suboptimal biochemical response were more likely to have adverse outcomes. Conclusion We constructed a nomogram, which can accurately predict the risk of inadequate biochemical response to UDCA, facilitating the early screening of high-risk patients with PBC who should be prioritized for additional therapy.

摘要： Co-presence of organic pollutants and heavy metals in soil is causing increasing concerns, but the lack of knowledge of relation between soil ecology and pollutant fate is limiting the developing of specific control strategy. This study investigated soil change under pyrene stress and its interaction with cadmium (Cd). Soil physicochemical properties were not seriously influenced. However, pollutants' presence easily varied soil microbial activity, quantity, and diversity. Under high-level pyrene, Cd presence contributed to soil indigenous microorganisms' adaption and soil microbial community structure stability. Soils with both pyrene and Cd presented 7.11-12.0% higher pyrene degradation compared with single pyrene treatment. High-throughput sequencing analysis indicated the proportion of Mycobacterium sp., a commonly known PAHs degrader, increased to 25.2-48.5% in treatments from 0.52% in control. This phenomenon was consistent with the increase of PAHs probable degraders (the ratio increased to 2.86-6.57% from 0.24% in control). Higher Cd bioavailability was also observed in soils with both pollutants than that with Cd alone. And Cd existence caused the elevation of Cd resistant bacterium Limnobacter sp. (increased to 12.2% in CdCK from 2.06% in control). Functional gene prediction also indicated that abundance of genes related to nutrient metabolism decreased dramatically with pollutants, while the abundances of energy metabolism, lipid metabolism, secondary metabolites biosynthesis-related genes increased (especially for aromatic compound degradation related genes). These results indicated the mutual effect and internal-interaction existed between pollutants and soils resulted in pollutants' fate and soil microbial changes, providing further information regarding pollutants dissipation and transformation under soil microbial response. 摘要： The aim of the present study was to evaluate the biochemical profile and reproductive performance in postpartum dairy cows with clinical metritis. The research was carried out using performed on 213 pluriparous lactating cows from two dairy herds in southeast Brazil. Dairy cows were assigned into a group of healthy cows (n = 147) and cows with metritis (n = 66). Blood samples to assess serum albumin, cholesterol, fibrinogen, calcium, gamma-glutammyltransferase (GGT), aspartate aminotransferase (AST) and beta-hydroxybutyrate (BHBA) concentrations were performed on day 8 +/- 2 postpartum. Metritic cows had lower (p < .05) serum albumin, cholesterol and calcium concentration, and higher (p < .05) GGT and AST activity, and BHBA and fibrinogen concentrations when compared to the healthy group. The calving-to-conception interval and services per pregnancy were higher in metritic cows (p < .01) than in healthy cows and the conception rate at first service was lower in metric group (p < .01). Variable importance in projection plots demonstrated that cholesterol was the main discriminator between metritic and healthy cows. Our results indicated alterations in the biochemical profile and impaired reproduction performance in metritic cows.

摘要： Introduction A number of guidance documents have been published in recent years for the diagnosis and management of hypogonadism (HG). Laboratory practice has a major role in supporting guidelines with accurate and precise serum total testosterone (TT) methods and standardised pre- and post-analytical protocols. Our study investigated whether laboratory practice currently supports the management guidelines for HG. Methods An internet-based questionnaire survey of senior laboratory biochemists (UK/Republic of Ireland) was conducted (April-May 2018). Questions reflected sampling, laboratory practice, reference ranges and reporting of results. The results were analysed in conjunction with data obtained from the UK National External Quality Assurance Service (UK NEQAS) on testosterone assay performance. Results Analyses of 96 laboratory surveys returned the following: 74 laboratories stated that the optimal sampling time was communicated to users;81 laboratories used immunoassays;76 laboratories included reference ranges for adult men (31 had dual/multiple age-related intervals). Wide variability in lower/upper limits was evident in the common immunoassays;the majority of reference ranges were from manufacturers (50.0%) or historical (18.8%). Action limits based on TT levels were used by 64 laboratories, but 63 did not report a borderline range as suggested by the guidelines. Protocols for cascading tests based on TT were evident in 58 laboratories, with 50 laboratories offering estimated free testosterone;interpretative comments were provided by 67 laboratories, but no references were made to the management guidelines. Data from UK NEQAS demonstrated considerable variation in testosterone assay performance. Conclusions Our survey has highlighted inconsistencies that could lead to HG (and other conditions requiring measurement of TT) not being managed appropriately. 摘要： Background: the ageing population has increased in many countries, including China. However, reference intervals (RIs) for older people are rarely established because of difficulties in selecting reference individuals. Here, we aimed to analyse the factors affecting biochemical analytes and establish RI and age-related RI models for biochemical analytes through mining real-world big data. Methods: data for 97,220 individuals downloaded from electronic health records were included. Three derived databases were established. The first database included 97,220 individuals and was used to build age-related RI models after identifying outliers by the Tukey method. The second database consisted of older people and was used to establish variation source models and RIs for biochemical analytes. Differences between older and younger people were compared using the third database. Results: sex was the main source of variation of biochemical analytes for older people in the variation source models. The distributions of creatinine and uric acid were significantly different in the RIs of biochemical analytes for older people established according to sex. Age-related RI models for biochemical analytes that were most affected by age were built and visualized, revealing various patterns of changes from the younger to older people. Conclusion: the study analysed the factors affecting biochemical analytes in older people. Moreover, RI and age-related RI models of biochemical analytes for older people were established to provide important insight into biological processes and to assist clinical use of various biochemical analytes to monitor the status of various diseases for older people.

摘要： Approaches to learning and teaching have been undergoing massive changes. Technology has enabled many innovations while other methods have embedded authentic research approaches or looked to other disciplines. The tools in education session of the conference looked at tools being used to teach biochemistry and molecular biology ranging from online platforms, authentic research experiences to the use of music.

摘要： Thiol-based redox regulation is a post-translational protein modification for controlling enzyme activity by switching oxidation/reduction states of Cys residues. In plant cells, numerous proteins involved in a wide range of biological systems have been suggested as the target of redox regulation;however, our knowledge on this issue is still incomplete. Here we report that 3-phosphoglycerate dehydrogenase (PGDH) is a novel redox-regulated protein. PGDH catalyzes the first committed step of Ser biosynthetic pathway in plastids. Using an affinity chromatography-based method, we found that PGDH physically interacts with thioredoxin (Trx), a key factor of redox regulation. The in vitro studies using recombinant proteins from Arabidopsis thaliana showed that a specific PGDH isoform, PGDH1, forms the intramolecular disulfide bond under nonreducing conditions, which lowers PGDH enzyme activity. MS and site-directed mutagenesis analyses allowed us to identify the redox-active Cys pair that is mainly involved in disulfide bond formation in PGDH1;this Cys pair is uniquely found in land plant PGDH. Furthermore, we revealed that some plastidial Trx subtypes support the reductive activation of PGDH1. The present data show previously uncharacterized regulatory mechanisms of PGDH and expand our understanding of the Trx-mediated redox-regulatory network in plants.
