摘要： Fluoride is the second largest contaminant of drinking water. Fluoride toxicity is a major concern in the endemic areas where a high amount of fluoride is present in ground water. Oxidative stress has been proposed to be one of the mechanisms of fluoride-induced toxicity. Antioxidant-rich food has been found to alleviate fluoride-induced toxicity. Therefore, in this study, we have examined the effect of hesperidin on fluoride-induced neurobehavioral changes in rats. In the current study, male Sprague-Dawley rats were exposed to sodium fluoride through drinking water (120 ppm). Hesperidin (200 mg kg(-1) d(-1);per os) was administered either alone or in combination with fluoride-containing drinking water. Bisphinol A diglycidyl ether (BADGE) was used as peroxisome proliferator-activated receptor-gamma (PPAR-gamma) antagonist and was administered (10 mg kg(-1) d(-1);intraperitoneal injection) with/without hesperidin along with fluoride-containing drinking water. The behavioral changes in the animals were assessed by analyzing rotarod test, novel object recognition test, and forced swim test (FST). After 8 weeks, animals were killed to isolate blood and brain for monitoring biochemical changes. The 8-week exposure of fluoride resulted in motor impairment as observed with reduced fall time in rotarod test, memory impairment as observed with reduced preference index in novel object recognition test, and depression-like behavior as observed with reduced mobility index in the FST. Treatment with hesperidin improved neurobehavioral impairment along with restoration in brain biochemical changes (ie, acetylcholinesterase activity and antioxidant and oxidative stress parameters). The protective effect of hesperidin was reversed by coadministration of BADGE. The neuroprotective effect of hesperidin appears to be contributed through PPAR-gamma receptor.

摘要： In this paper, we reveal and characterize cross-feeding behaviour between the common gut commensal Bacteroides cellulosilyticus (Baccell) and certain bifidobacterial strains, including Bifidobacterium breve UCC2003, when grown on a medium containing Larch Wood Arabinogalactan (LW-AG). We furthermore show that cross-feeding is dependent on the release of beta-1,3-galacto-di/trisaccharides (beta-1,3-GOS), and identified that the bga gene cluster of B. breve UCC2003 allows beta-1,3-GOS metabolism. The product of bgaB is presumed to be responsible for the import of beta-1,3-GOS, while the bgaA gene product, a glycoside hydrolase family 2 member, was shown to hydrolyse both beta-1,3-galactobiose and beta-1,3-galactotriose into galactose monomers. This study advances our understanding of strain-specific syntrophic interactions between two glycan degraders in the human gut in the presence of AG-type dietary polysaccharides.

摘要： A novel lipolytic enzyme-encoding gene,lipO745, fromAspergillus oryzaeRIB40 was cloned and expressed inPichia pastoris. Purified recombinant LipO745 (rLipO745) had a molecular mass of approximately 60 kDa on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. rLipO745 exhibited maximum activity at 40 degrees C and pH 7.0 and was stable at temperatures <= 40 degrees C. The substrate specificity of purified rLipO745 was analyzed using alpha-naphthyl esters as artificial substrates and various triacylglycerol and sterol esters as natural substrates. From among a panel of alpha-naphthyl esters (C2-C16), alpha-naphthyl butyrate (C4), with an activity of 269 +/- 3.3 units/mg protein, was the optimal substrate for hydrolysis by the purified recombinant protein. TheK(m)andk(cat)values of rLiO745 for the C4 substrate were 0.073 +/- 0.0012 mM and 608 +/- 108 s(-1), respectively. The purified recombinant enzyme had considerable hydrolytic activity toward tributyrin, tripalmitin, and triolein, indicating lipase activity, and toward cholesteryl acetate, butyrate, palmitate, and oleate, indicating sterol esterase activity. Transesterification activities between tributyrin and cholesterol or between tributyrin and campesterol were also determined.

摘要： Glandless cotton can be grown to obtain cotton seeds free of toxic gossypol for use as both food and feed. However, they are not grown normally due to their lesser productivity and higher susceptibility to biotic stress. Great attention has been paid to biotic stresses rather than abiotic stresses on glandless cotton. Chromium (Cr) is a common pollutant of soil and considered a serious threat to plants due to its adverse effects on different functions. Although numerous studies are available on the toxicity of Cr6+ in various plants. However, its adverse effects and mechanism of toxicity in glandless cotton can seldom be found in the literature. This study examined the Cr6+ effect on glandless cotton in comparison to glanded cotton. Four pairs of glanded and glandless cotton near-isogenic lines (NILs) were exposed to different doses (0, 10, 50, and 100 mM/L) of Cr6+ for seven days, and biochemical, physiological, molecular, and ultrastructure changes were observed, which were significantly affected by Cr6+ at high concentrations in all NILs. The effect of Cr6+ on ionic contents shows the same trend in glanded and glandless NILs except for manganese (Mn2+) that show inhibition in glandless (ZMS-12w and Coker-312w) and enhance in the glanded NIL (ZMS-17). The gene expression of superoxide dismutase (SOD) and peroxidase (POD) revealed similar trends as enzyme activities in glandless NILs. The principal component analysis (PCA) and Agglomerative hierarchical clustering (AHC) results of all NILs from morpho-physiological traits, cluster ZMS-16, and ZMS-17 into Cr6+ sensitive group. While the glandless NILs have the potential to cope with the Cr toxicity by increasing the antioxidant enzyme activity and their gene expression. This study also revealed that Cr6+ tolerance in cotton is genotypic and has an independent mechanism in the root that not related to low gossypol. (C) 2020 Elsevier Ltd. All rights reserved.

摘要： The study, which covers the period between 2014 and 2018, was carried out in the city of Naberezhnye Chelny, Republic of Tatarstan, Russia. The aim of the study was to examine the biochemical response of maple trees growing in the anthropogenic environments. Leaf samples from 600 trees (Acer platanoides L. and Acer negundo L.) were collected at monthly intervals from June through August. Sampling was performed early in the morning (11 a.m.) in the middle of the month. The study offers statistical data on the tannin content, determined via permanganometry;the ascorbic acid concentration, found via titration with 2.6-dichlorophenolindophenol;the ascorbate oxidase activity determined by absorbance at 265 nm;and the polyphenol oxidase activity, found by the spectrophotometric method. Relatively higher ascorbate oxidase activity was detected in August among ash-leaved Acer platanoides L. and Acer negundo L. in areas with strong anthropogenic impact. Due to increased air pollution, maple trees were found to exhibit an increase of polyphenol oxidase activities. The condensed tannin content in Norway maple trees dropped over time: by 1.24 in July (avenue);by 0.94 (buffer area) and 0.76 (avenue) in August. The condensed tannin content in the ash-leaved maple trees also decreased: by 0.69 (buffer area) and 0.22 (avenue) in July;by 0.37 (buffer area) and 0.61(avenue) in August. (C) 2020 The Authors. 摘要： Sub-lethal toxic impacts of chromium on hematological, biochemical and histological parameters were analyzed in the female small Indian mongoose (Urva auropuctatus) residing contaminated environment of tannery industry. Chromium bioaccumulation in the blood, liver and kidney tissue of the exposed mongooses was found elevated compared to the control mongooses' tissues. Total body weight (75.7%), liver weight (83.6%) as well as HSI (68.1%), RSI (86.2%) and the platelets counts (59.7%) were found significantly elevated, with significantly reduced RBCs (59.6%), and WBCs (64%). LFT and RFT were also found abnormal, moreover, the histopathological injuries had been distinct inside the kidney (>75%) and hepatic (>75%) tissues of exposed animals. Shrinkage and vacuolization (> 75%) inside the hepatocyte expanded sinusoidal spaces and nuclear pyknosis (>75%) was evident within the hepatic tissue. Hypertrophy of epithelial cells of renal tubules and inter-renal cells of the head kidney with a reduction in tubular lumens (>75%) and vacuolization of tubules were witnessed within the kidney section. Atrophy inside the kidney inter-renal cells, glomeruli compression within the Bowman's capsules (>75%) following the necrosis in hematopoietic tissues were found in exposed animals. The present findings indicate that chronic exposure to chromium induces severe anemia, decreased serum protein concentration, hepatic and renal tissue histopathology, impairing the vital capabilities of liver, metabolic regulation, excretion, and stress homeostasis maintenance of which within the long-run may posture a severe risk to animal well-being then distress their inhabitants. 摘要： Bivalve stocks have been decreasing in the last decades largely due to emergent diseases and consequent mass mortality episodes. Cerastoderma edule (the edible cockle) is one of the most exploited bivalves in Europe and is among the most common hosts for trematodes, the most prevalent macroparasites in coastal waters but yet poorly studied. Therefore, in the present study, this bivalve species was used as host model to determine if trematode infection exerts a negative effect on bivalve energy metabolism and balance and if the tissues targeted by different trematodes influence the metabolic cost, with physiological and biochemical consequences. Cockles were experimentally infected with two trematode species, Himasthla elongata and Renicola roscovitus, that infect the foot and palps, respectively. Trematode infection exerted a negative effect on the metabolism of C. edule, the second intermediate host, by reduction of oxygen consumption. A different host biochemical response was found depending on trematode species, especially in regard to the level of oxygen consumption decrease and the preferential accumulation of lipids and glycogen. This study represents a step towards the understanding of hosttrematode relationships that can be used to better predict potential conservation threats to bivalve populations and to maximize the success of stock and disease management.

摘要： In many health-related programs biochemistry and molecular biology are core subjects, but these subjects are often not the students main focus. This challenges educators to develop curriculum that demonstrates the relevance of biochemistry and molecular biology and engages these students. This conference session discussed the value of biochemistry and molecular biology education in the health sciences and the methodologies which can be implemented.

摘要： The release of pharmaceutical chemicals in the biosphere can have unpredictable ecological consequences, and knowledge concerning their putative toxic effects is still scarce. One example of a widely used pharmaceutical that is present in the aquatic environment is ciprofloxacin. Previous indications suggest that this drug may exert several adverse effects on exposed biota, but the characterization of a full ecotoxicological response to this drug is far from complete, especially in estuarine ecosystems. This work aimed to characterize the acute and chronic effects of ciprofloxacin in the polychaete Hediste diversicolor (Annelida: Polychaeta), exposed to environmentally relevant levels of this drug, close to the real concentrations of this pharmaceutical in surface waters. The adopted toxic endpoints were behavioral parameters, combined with a biomarker-based approach (quantification of the activities of catalase (CAT), glutathione-S-transferase (GSTs), cholinesterases (ChEs), glutathione peroxidase (GPx), and lipid peroxidation levels. Exposure to ciprofloxacin caused effects on behavioural traits, such as an increase in burrowing times and hyperactivity, alongside alterations in biomarkers, including a significant increase in CAT activity following acute exposure. In addition, and after both acute and chronic exposure, lipid peroxidation was reduced, while AChE activities were enhanced. It was possible to ascertain the occurrence of pro-oxidative alterations following exposure to low levels of ciprofloxacin, which were counteracted by the triggering of CAT activity. The meaning of the enhancement of AChE activity is not clear, but it appears to be linked with the observed behavioural changes, and may have been associated with the stimulation of the behavioural traits. These data strongly suggest that the presence of ciprofloxacin in estuarine areas is not without risks, and exposed biota, namely polychaete species, are likely to have their ecological roles affected
