This study aimed to investigate the effects of Plantago asiatica polysaccharide (PLP) on the growth performance, organ development, immune function and cecal microbiota of yellow-feathered broilers. Four hundred 1-day-old healthy Daheng broiler roosters were randomly divided into 2 groups with 10 replicates per group and 20 broilers per replicate. Broilers in control group were fed a basal diet, and those in PLP group were fed the basal diet supplemented with 1% PLP. The feeding experiment lasted for 49 days. At 21 and 49 days of age, samples were collected for immune organ indexes, intestinal morphology, and serum immune indexes determination. At 49 days of age, cecal contents were collected for metagenomic sequencing analysis. The results showed as follows: 1) compared with control group, the final body weight, average daily gain and average daily feed intake of broilers in the PLP group were significantly increased (P<0.05), and the increased ( P<0.05), and the ratio of feed to gain was significantly reduced ( P<0.05). 2) At 21 days of age, the Fabricius index, duodenal villus height and villus height/crypt depth ratio in the PLP group were significantly higher than those in the control group ( P<0.05);at 49 days of age, the duodenal villi height in the PLP group was significantly higher than that in the control group (P<0.05). 3) The contents of serum total protein (TP) (21 and 49 days of age), immunoglobulin A (IgA) (21 and 49 days of age) and immunoglobulin G (IgG) (49 days of age) in the PLP group were significantly higher than those in the control group (P<0.05). 4) The metagenomic sequencing results showed no significant differences in ct diversity of cecal microbiota between control group and PLP group (P>0.05);compared with the control group, the relative abundances of cecal Clostridium, Anaeromassilibacillus, Roseburia, Eisenbergiella and BittareUa in the PLP group were significantly increased (P<0.05). 5) KEGG results showed that there was no significan
